The cytotoxic effects of EGCG on Huh7 cells were evaluated by MTT assay. The expression of HCV structural and nonstructural proteins was detected by Western blotting. The EGCG-induced activation of RNase L was confirmed by an RNase L activity assay. We found that EGCG inhibited HCV replication in a dose-dependent manner. EGCG also induced the activation of RNase L in Huh7 cells in a dose-dependent manner. However, it did not induce significant changes in the expression of HCV core and NS5A proteins. EGCG at a concentration of 100 microg/ml was the most efficient in suppressing HCV replication. Therefore, in this study, we further determined the effective inhibitory concentration (EC(50)) of EGCG and found that it was 20 microg/ml, which was much lower than the concentration (100 microg/ml) of EGCG that maximally activated the RNase L/p56(lck) pathway in Huh7 cells.
